- APPEL A PROJETS EN 2008 DE LA FONDATION DE RECHERCHE ALLIANCE BIOSECURE
La Fondation de recherche Alliance BioSecure reconnue d'utilité publique en décembre 2006, a pour objectifs de contribuer à garantir la sécurité biologique dans les domaines de la santé et de l'alimentation. Elle renouvèle son Appel à Projets en 2008 pour initier des projets de recherche innovants répondant aux thématiques de recherche sélectionnées par son Conseil Scientifique. Il y a quatre thématiques de recherche centrées sur les maladies liées à un repliement anormal de la protéine cible, par exemple les maladies à prions :
I : Compréhension du processus de repliement anormal, caractérisation des mécanismes induisant ce repliement anormal, analyse des interactions entre la protéine anormale et ses partenaires, liens physiopathologiques entre la protéine anormale et les symptômes cliniques.
II : Recherche, développement ou validation de méthodes de détection des prions (et autres protéines à repliement anormal) dans les liquides biologiques ou organes humains et animaux.
III : Méthodes d'élimination ou d'inactivation des contaminants potentiels dans la production de produits biologiques et dans le nettoyage des instruments et des surfaces.
IV : Surveillance et épidémiologie des maladies à prion.
Les projets ne concernant pas les prions sont éligibles, dans la mesure où ils répondent à des problématiques actuelles de santé publique.

- Post-doctoral position on “Innate Immune signaling in intestinal homeostasis and defence towards enteropathogens”
Project: Our laboratory is affiliated to Inserm. We are interested in elucidating the critical processes involved in both engagement of physiological and pathological innate immune signaling in regards to tissue homeostasis and host defence towards certain enteropathogens. We have recently shown that Nod2, a molecule sensing bacterial muramyl dipeptide, regulates intestinal defence towards bacterial infection and play a major role in Crohn’s disease development. A post-doctoral position is available in our laboratory to further dissect the regulation of innate immune signaling in the intestinal mucosa and related barrier organs. The successful candidate will use and develop novel mouse models and appropriate cell-based assays.
Candidate profile: PhD and/or MD Candidates with outstanding expertise in molecular/cellular biology, antimicrobial immunology and/or microbiology. Fluent knowledge of the English and skills in pathology and mouse experiments are mandatory. A two-year Post-doctoral fellowship position is available at the Pierre & Marie Curie University (Paris VI), in the “Immunology-Immunopathology-Immunotherapy Unit” (I3, UMR7087). Located in the Pitié-Salpêtrière Hospital, our group is well established and is developing new genetic vaccines strategies. The team is composed of 1 researcher, 1 post-doctorant, 2 Ph-D students, 2 engineers and 1 technician.
The post-doc fellow will be financed by a European grant; the general objective of this grant is to generate multi-step improved and efficient DNA-based therapeutic vaccine against HIV.
This project focuses specifically on optimization of the “plasmoVLP” strategy and application for HIV vaccine development.
“PlasmoVLP” combines the advantages of virus-like particles (VLPs) and DNA vaccines by engineering them into single vectors, i.e. by designing plasmids that harbour the genetic code necessary for production of recombinant VLPs. This has been achieved using murine leukaemia virus-derived system allowing expression of antigens within and/or onto retrovirus-like particles. We previously demonstrated that “plasmoVLP” represent a significant improvement in DNA vaccination (Bellier et al, Vaccine 2006; Dalba et al, Mol Therapy 2007).
We now propose to modify the retroviral constructs to: (i) incorporate “plasmoVLP” cassette into viral vectors (adenovirus, measle …), (ii) modify epitope insertion sites (into gag and/or env), (iii) add immunostimulatory molecules within/onto VLPs and apply all these optimizations for HIV vaccination.
REQUIREMENT:
Applicants must be trained in molecular biology and virology. An experience in retrovirology, immunology or vectorology would be a plus.
Successful applicant should be highly motivated, with good presentation and communication skills, and able to work in a team environment.

L’Institut Pasteur vient d’inaugurer son « Centre d’Immunologie Humaine (CIH)», dédié à la recherche dite translationnelle : celle-ci vise à favoriser l’application de découvertes issues de la recherche fondamentale aux domaines du diagnostic, du traitement ou de la prévention des maladies humaines. Les travaux du CIH seront particulièrement axés sur l’immunothérapie des tumeurs et l’immunopathogenèse des cancers, les maladies infectieuses (relations hôtes/pathogènes), les maladies autoimmunes et les réactions allergiques.
